図書館法による「司書（補）」の資格を得るには、大学で司書資格取得に必要な科目を履修するか司書講習を受ける必要がある。通信制で学べる大学もある。
公立図書館や公立大学の司書になるには、地方公務員試験に合格する必要がある。この際、最初から司書（補）の資格を前提にして専門試験を行い図書館に採用する場合と、地方公務員行政職の試験を受けて採用され、図書館に配属される場合がある。また、公立図書館の場合、地方公共団体から図書館業務を受託している企業に就職して図書館の司書として勤務するケースもある。
学校図書館で学校司書として働くには、各地方公共団体の定める、司書資格や司書教諭資格、教諭免許状、相当実務経験等の資格要件を満たしたうえで学校司書の募集に応募して採用される必要がある。なお、学校図書館の司書教諭には主幹教諭、指導教諭又は教諭が充てられる。
国立大学図書館の場合は、大学法人が行う採用試験、私立大学では職員の採用試験を受ける。司書の仕事を行う上でまず必要なのは図書館情報学の知識であり、コンピュータや外国語に強ければ一層有利である。広範で多量の資料に目を通し、正確に分類・記録するには、知的好奇心や記憶力、反復して分類作業を行う持久力も必要である。
