「完成検査」は、自動車の安全性を確保するために重要な工程であり、その時点において自動車が道路を安全に走行でき、環境への基準を満足しているかを確認することであり、検査を適切に行うことによって、整備不良や不正改造箇所のある自動車が公道を走行することを防いでいる。もし完成検査をした車に不具合があれば、人命に関わる事故の原因となりうるため、完成検査は継続検査（車検）の中で最も責任感が問われる重要な仕事である。
具体的には、自らの指定整備工場で整備された自動車について、構造に関する検査として、車検証に記載されている寸法との同一性や最低地上高等を視認等により検査する。装置に関する検査として、制動装置（ブレーキ）の性能及び制動能力や前照灯（ヘッドライト）の明るさ及び主光軸の向き等を検査機器を用いて基準値内であるかを検査するほか、動力伝達装置、走行装置、制動装置、灯火装置等の自動車の各装置について、検査用ハンマーを用いる方法や目視等により検査する。
継続検査の場合、一連の工程を見ると、車検用車両の受け入れ、整備における作業指示、計測機械等を用いた完成検査、指定整備記録簿への検査結果記載、保安基準適合証への証明を行う書類作成、車検依頼者への報告という流れが一般的である。使用者に交付する記録簿には、測定結果、消耗品の残量等、点検結果、整備の内容などが記載され、自動車検査員が、車両の改善提案やメンテナンスの助言を行うこともある。
指定整備工場における完成検査は、１台あたり数十分程度の所要時間である。検査工場の規模にもよるが自動車検査員１人あたり１日に４～５台、多くても８台ほどである場合が多い。普通乗用車の場合、所有者への引き渡しまで１日、トラック等で２～３日というのが一般的である。
また、指定整備工場における完成検査として扱える車種は、指定を受けた自動車の種類に限られるものであるが、実際には勤務している現場が取り扱っている車種のみを日常的には検査するというのが一般的となる。
働く場所が同じであることから混同されることもある自動車整備士は、定期点検や車検などにおいて、不具合・故障箇所がないかを点検し、それらがあった場合の修理・整備などメンテナンスを行うことが主な業務である。他方、自動車検査員は自動車整備士が行った整備に不備・漏れがないか、整備後の自動車が国の基準に適合し問題なく安全に走行できるかについて検査と確認をする。言い換えるならば、自動車整備士は自動車の整備全般を行い、車検に合格するよう点検と整備をするが、車検における完成検査を行うことはできず、自動車検査員による最終判断が必要となる。また、自動車検査員は、整備と検査を同一の者が行うことはできない。
自社工場で車検を行うことができる指定整備工場においては、自動車検査員を最低でも１名を配置する必要がある。自動車検査員は、現場において、資格を持たない工員や自動車整備士の監督業務を行うこともあることから、自動車整備士の仕事にも深い理解が必要である。
自動車検査員は、人の命に関わる大きな責任を伴う仕事を行うことから、まずは自動車に関する豊富な知識を持つことが大前提となる。細部に亘る項目の検査をするためには、曖昧な知識では通用せず、自動車検査員の選任を受けた後も、自動車の基本構造から最新の技術やシステムの動向等、常に新しい知識を吸収し続ける必要がある。その上で、「安心・安全な車を社会に届けたい」といった強い責任感を持って仕事に取り組むことができる者が向いている。また、日常の作業動作としては、微細な異状を察知する力や、集中力があり繰り返しの同じ作業が苦にならない者が向いている。

自動車検査員教習の受験資格は、整備主任者としての実務経験が二級自動車整備士の場合は１年以上、一級自動車整備士の場合は６ヶ月以上必要であること（整備主任者になるには、二級自動車整備士又は一級自動車整備士の資格が必要）、直近の整備主任者研修を受講修了していること、勤務先が「指定整備工場」であることなどである。
