通関士になるには国（財務省・税関）が行う通関士試験に合格しなければならない。試験は実務経験があると一部免除されるため、通関業者に就職してから会社内や通関業者団体で行っている研修会、通信教育で勉強し、通関士の資格獲得を目指す人も多い。ただし、学歴、年齢、経歴、国籍等についての受験資格制限はない。
試験では、通関業法、関税法・関税定率法、外国為替及び外国貿易法などの関係法令に熟知する他に、輸出入申告書、輸入（納税）申告書などの通関手続に関する書類作成についての十分な知識が求められる。また、通関書類の付属書類である仕入書（インボイス）、包装明細書（パッキングリスト）、輸出入承認書、原産地証明書などについても十分な理解が求められる。
通関業務で扱う書類は英語で書かれているので、英語力も必要である。
なお、通関士試験に合格したものは、受験地に関わりなく、どの税関の管轄区域においても通関士となることができる。
