入職にあたって、特に学歴や資格は必要とされないが任用資格として都道府県の認定する「放課後児童支援員」があり、おおむね児童40人につき2名以上の同認定者を配置することが定められている。
放課後児童支援員の任用資格の要件は、保育士・社会福祉士・教員免許などの有資格者、大学で社会福祉学等の一定の課程を履修した者のほか、5年以上（高卒以上相当の学歴の者は2年以上）の関連する実務経験があり市町村長が適当と認めた者なども含まれているため、入職後に取得を目指すことも可能である。
要件を満たしている者は都道府県が実施する16科目24時間の「放課後児童支援員認定資格研修」を受講、修了することで資格を取得できる。その際、保育士など特定の資格保持者は一部の研修が免除となる。
なお、学童保育の事業者に対して放課後児童支援員の資格保持者の人員配置を「2名以上」と義務付ける全国統一基準が示されていたが、人材確保が難しいとの意見を受け、2020年度以降は市町村に人員配置規定の取り扱いが委ねられることとなっている。
学童保育指導員は子どもの生活と直接的にかかわるため、仕事の遂行にあたって人権やプライバシーの尊重、児童・保護者との信頼関係の構築、保育関連の専門性が求められる。また、業務遂行にあたって同僚とのチームワーク、必要に応じ学校、地域の関係者との連携が重要であり協調性やコミュニケーション力も必要である。更に最近では、外国籍の子どもや保護者が増えていることから、外国語の日常的な会話能力が求められるケースもある。
