入職にあたって、特に学歴や資格は必要とされない。杜氏、蔵人には、特定の地域出身の就業者が多いという特色から、入職経路は縁故により杜氏集団に入ることが多い。入職すると、最初は蒸米や酒母などの各部署の補助につき、少し経験を積むと各部署を担当する係員となる。その上には作業を統括する役として三役（もろみ主任、麹主任、酒母主任）があり、杜氏を補佐して毎日の作業内容の指示、管理などを行う。全体の責任者が杜氏であり、規模の大きいところでは杜氏補佐を置く場合もある。
蒸米・麹・酒母・もろみの適否の判断など、人間の勘や経験によるところが多いため、経験や熟練に基づく判断力、観察力や洞察力が必要となる。
関連する資格に厚生労働省の技能検定「酒造技能士」がある。
清酒製造では多くは数カ月の間、寝食をともにし、様々な工程で協力して作業をするので、協調性や自分から進んで作業をする姿勢も求められる。
杜氏集団に組み込まれて入職する以外に、清酒メーカーに雇用されるルートもある。
